Epithalon vs Humanin

Epithalon and humanin both appear in discussions of ageing biology, but one is a synthetic tetrapeptide and the other is a naturally occurring peptide that the body makes 1,12. Epithalon (epitalon, Ala-Glu-Asp-Gly) was synthesised from the amino-acid content of a bovine pineal extract, and in human cell culture it induced telomerase activity 1,3. Humanin is a 24-amino-acid mitochondrial-derived peptide first identified in 2001 in a screen for factors that protect neurons from Alzheimer's disease-related insults 5,8,12. Neither has been tested as a treatment in a published controlled human trial in the sources reviewed, and neither is FDA-approved 1,13.

FieldEpithalonHumanin
ClassSynthetic tetrapeptide (Ala-Glu-Asp-Gly) 1Endogenous 24-amino-acid peptide; classed as a mitochondrial-derived peptide 8,12
Origin and structureSynthesised from the amino-acid composition of epithalamin, a bovine pineal gland extract; the FDA treats the two as different substances 1,4Identified in 2001 through functional expression screening of a cDNA that rescued neuronal cells from Alzheimer's disease gene-induced death 5
Proposed mechanismInduced telomerase expression and telomere elongation in human fetal fibroblasts 3; reported to raise melatonin levels by an unknown mechanism 1In cell studies, binds the pro-apoptotic proteins Bax and Bid 7 and IGFBP-3 6,8; acts through a receptor complex involving CNTFR-alpha, WSX-1 and gp130 8
Half-lifeNo pharmacokinetic data found by the FDA 1Not located in the sources reviewed
Highest human trial phaseNo completed trials identified; one early phase 1 multi-agent ageing study (NCT07888595) matched a registry search 17No interventional trials; registered human studies measure humanin as a blood biomarker only 16
FDA statusNot FDA-approved; the FDA proposed it not be added to the 503A compounding bulks list (PCAC, 24 July 2026) 1,2; previously listed among withdrawn category 2 nominations 18Not FDA-approved; no Drugs@FDA record (queried 24 October 2026) 13; not in any 503A nomination category as updated 14 May 2026 14
Published human outcomesThe FDA found no studies in people with insomnia; human reports concern melatonin levels in older people, mostly with epithalamin 1Observational only: lower circulating levels with age 9, lower levels in coronary endothelial dysfunction 10, and a rise after endurance exercise 11
Safety signalsFDA found no human safety data and flagged a theoretical cancer concern because it activates telomerase 1No human dosing studies located, so no safety data in the sources reviewed 16
WADA statusNot named on the 2026 Prohibited List; S0 covers substances with no governmental approval for human use 15Not named on the 2026 Prohibited List; S0 covers substances with no governmental approval for human use 15
Who funded the evidenceLargely the St Petersburg group of Vladimir Khavinson and the Russian Academy of Medical Sciences 1,3Academic groups in Japan, the United States and Sweden; one human study was funded by Swedish regional and kidney foundations 5,9,11

Epithalon: what the data says

Epithalon was synthesised from the amino-acid composition of epithalamin, a bovine pineal extract, and most studies come from the developer's research network 1,4. In human fetal fibroblast cultures (cell data), it induced the catalytic subunit of telomerase, telomerase activity and telomere elongation 3. A 2025 review summarises cell, animal and computational studies reporting effects on melatonin synthesis and several enzymes, while noting that its mechanisms remain uncertain 4.

The FDA's 2026 review found no studies of epitalon in people with insomnia, no pharmacokinetic data and no human safety data 1. It judged available mouse toxicology studies too limited to inform cancer risk, a gap it considered relevant because epitalon activates telomerase 1.

Humanin: what the data says

Humanin was identified in 2001 as a secreted polypeptide that, in cell culture, prevented neuronal death caused by several familial Alzheimer's disease genes and amyloid-beta 5,6. In cell studies it binds pro-apoptotic Bcl-2 family proteins such as Bid 7 and the binding protein IGFBP-3 6,8. In rodents (animal data), brain infusion of humanin and intravenous infusion of analogues improved insulin sensitivity 9.

Human data are observational and concern the body's own humanin. Circulating levels were lower with age in humans and mice 9, lower in 20 patients with coronary endothelial dysfunction than in 20 without 10, and rose after acute endurance exercise but not resistance exercise in a 30-person study 11. No interventional human trials were found 16.

Key research differences

  • Origin: Epithalon is a synthetic peptide modelled on a pineal extract 1, whereas humanin is produced naturally and classed as a mitochondrial-derived peptide 12.
  • Size: Epithalon has four amino acids 1, while humanin has 24 8.
  • Proposed mechanism: Epithalon work centres on telomerase and melatonin 1,3, whereas humanin work centres on blocking apoptosis proteins and a cytokine-type receptor complex 7,8.
  • Human data: for Epithalon, the FDA found no insomnia studies 1; for humanin, human studies measure natural blood levels rather than test it as a treatment 10,11.
  • FDA review: the FDA formally reviewed Epithalon for compounding in 2026 1,2, while humanin does not appear in any 503A nomination category 14.

Where the evidence is limited

No study has compared Epithalon and humanin. Epithalon evidence is mainly cell and animal work from one research network 1,4. Humanin human evidence is correlational, measuring natural blood levels, and does not show what giving the peptide would do 9,10,11. Animal studies of humanin often used modified analogues rather than the native peptide 9.

Frequently asked questions

Are Epithalon and humanin both made by the body?
Humanin is a naturally occurring peptide 5,12. Epithalon is synthetic, modelled on the amino-acid content of a bovine pineal extract 1.
Has either been tested as a treatment in humans?
No controlled treatment trials were found. Registered humanin studies measure blood levels only 16, and the FDA found no Epithalon insomnia studies 1.
Can Epithalon and humanin be compared directly?
Not on current evidence. They have different structures and proposed mechanisms, and no study has tested them side by side 1,8.
What does the FDA say about Epithalon?
In a July 2026 briefing document, the FDA found no human safety data, raised a theoretical cancer concern related to telomerase activation, and proposed that it not be added to the 503A bulks list 1,2.
