Epithalon vs Pinealon

Epithalon and Pinealon are both short synthetic peptides from the same Russian research programme on pineal-gland peptides, led by Vladimir Khavinson in St Petersburg 1,5,7. Epithalon (Ala-Glu-Asp-Gly) was synthesised from the amino-acid content of a bovine pineal extract and in human cell culture induced telomerase activity 1,3. Pinealon (Glu-Asp-Arg, also called the EDR peptide) was studied for effects on reactive oxygen species in nerve and other cells 5,7. The FDA reviewed Epithalon for compounding in July 2026 and proposed it not be listed; Pinealon does not appear in the FDA's compounding nomination categories at all 1,11.

Epithalon: what the data says

Epithalon was synthesised from the amino-acid composition of epithalamin, a bovine pineal extract, and most of its literature comes from the group that developed it 1,4. In human fetal fibroblast cultures (cell data), it induced the catalytic subunit of telomerase, telomerase activity and telomere elongation 3. In fluorescence studies in HeLa cells (cell data), labelled Epithalon entered the nucleus and bound preferentially to DNA sequences containing CNG motifs 9. In organotypic rat pineal cultures, it stimulated the proliferation marker Ki-67 and the transcription factor CGRP 8.

The FDA's 2026 review found no studies of epitalon in people with insomnia, no pharmacokinetic data and no human safety data, and judged mouse toxicology too limited to inform cancer risk 1.

Pinealon: what the data says

Pinealon (Glu-Asp-Arg) was studied mainly in cells. In cerebellar granule cells, neutrophils and PC12 cells (cell data), it limited reactive oxygen species accumulation and necrotic cell death, with delayed ERK1/2 activation and cell-cycle changes; the authors proposed direct interaction with the genome 5. In rats whose mothers were given a methionine load during pregnancy (animal data), offspring given Pinealon showed changes in spatial-orientation and learning measures and lower reactive oxygen species in isolated cerebellar neurons 6. A 2020 review by the same group summarises reported effects on signalling pathways and antioxidant-enzyme synthesis 7.

Human evidence is a single small Russian-language study in 32 people aged 41 to 83 with chronic multimorbidity and an organic brain syndrome in remission, which reported changes in biological-age indicators and, alongside them, prooxidant activity on chemiluminescence and lower CD34+ cell markers in blood 10. No registered trials were found 13.

Key research differences

  • Structure: Epithalon is a four-amino-acid peptide 1, while Pinealon is a three-amino-acid peptide 5.
  • Proposed mechanism: Epithalon work centres on telomerase and melatonin 1,3, whereas Pinealon work centres on limiting reactive oxygen species and on proposed DNA binding 5,7.
  • Human data: Epithalon human reports concern melatonin levels, often with the related extract epithalamin 1; Pinealon has one small Russian study of biological-age markers 10.
  • Regulatory attention: the FDA formally reviewed Epithalon for compounding in 2026 1,2, while Pinealon does not appear in any 503A nomination category 11.
  • Reported redox direction: cell studies describe Pinealon as limiting reactive oxygen species 5, yet the 32-person human study reported prooxidant activity on chemiluminescence 10.

Where the evidence is limited

No study has compared Epithalon and Pinealon in people. Both come from one research network, which limits independent replication, and several papers test them side by side only in cell cultures 7,8,9. The single human Pinealon study was small, Russian-language, used composite biological-age measures and combined the peptide with another, so its findings cannot be attributed to Pinealon alone 10. For Epithalon, the FDA found no clinical insomnia data, no pharmacokinetics and no human safety data 1.

Frequently asked questions

Are Epithalon and Pinealon related?
Both are short synthetic peptides from the same St Petersburg pineal-peptide research programme, but they have different sequences: Ala-Glu-Asp-Gly and Glu-Asp-Arg 1,5.
Is Pinealon the same as the EDR peptide?
Yes. Reviews from the developing group use EDR (Glu-Asp-Arg) and Pinealon for the same tripeptide 5,7.
Has either been tested in a registered clinical trial?
No completed trial of either was identified. A registry search returned no Pinealon studies and, for epitalon, one early phase 1 multi-agent ageing study 12,13.
Can Epithalon and Pinealon be compared directly?
Only in cell culture. Several papers test both in the same cell systems 8,9, but no human study compares them 1,10.
