Kisspeptin-10 vs Kisspeptin-54
Kisspeptin-10 and kisspeptin-54 are two forms of the same peptide system, differing in length and in how far their human evidence has gone 1,2. Kisspeptin-54 is described as the major circulating isoform of kisspeptin in humans, and it has been taken as far as phase 2 trials to trigger egg maturation in IVF 2,6. Kisspeptin-10 is the shortest fragment retaining full activity at the receptor, with human evidence limited to hormone-response studies in small groups of healthy men 1,3. Neither is FDA-approved, and the WADA 2026 list names kisspeptin and its agonist analogues under testosterone-stimulating peptides in males 9,12.

Kisspeptin-10: what the data says
Kisspeptin-10 is the shortest kisspeptin sequence that retains full activity at the kisspeptin receptor 1. In healthy men, intravenous boluses produced a rapid, dose-related rise in serum LH, and continuous infusion raised LH pulse frequency, pulse size and testosterone 1.
A 2026 randomised, single-blinded, placebo-controlled study in 15 healthy men examined longer administration: continuous subcutaneous infusion over 5 days left gonadotropins similar to vehicle while testosterone stayed raised, whereas daily intermittent 8-hour infusions over 12 days sustained gonadotropin increases, and a later bolus still produced a response, indicating the receptor remained functional 3. Registered kisspeptin-10 studies are small and single-centre 10, and the FDA lists kisspeptin-10 in 503A category 2 11,13.
Kisspeptin-54: what the data says
Kisspeptin-54 is described as the major circulating kisspeptin isoform in humans, with a measured plasma half-life of about 28 minutes in male volunteers 2. In 6 men, infusion raised LH, FSH and testosterone versus saline 2. In women, a subcutaneous bolus raised LH in all phases of the menstrual cycle, with the largest rise in the preovulatory phase 4.
It has gone further clinically. In 53 women undergoing IVF, a single injection induced egg maturation, with fertilisation and embryo transfer in 92% and clinical pregnancy in 23% 8. In a phase 2 open-label randomised trial in 60 women at high risk of ovarian hyperstimulation syndrome, egg maturation occurred in 95%, live birth per transfer was 45%, and no woman developed moderate, severe or critical ovarian hyperstimulation syndrome 6. Repeated twice-daily injection over 2 weeks in women with hypothalamic amenorrhoea produced a markedly reduced hormone response by day 14, described by the authors as desensitisation 5. A 2025 randomised crossover study reported that intranasal kisspeptin-54 raised LH in healthy men and women and in patients with hypothalamic amenorrhoea 7.
Key research differences
- Length: kisspeptin-10 has 10 amino acids, kisspeptin-54 has 54 and is the major circulating human isoform 1,2.
- Pharmacokinetics: a plasma half-life of about 28 minutes has been measured for kisspeptin-54 in men 2; no half-life was located for kisspeptin-10.
- Clinical reach: kisspeptin-54 has phase 2 IVF trials with egg maturation and live-birth outcomes 6, whereas kisspeptin-10 evidence is hormone responses in small groups of healthy men 1,3.
- Repeated administration: twice-daily kisspeptin-54 over 2 weeks produced desensitisation 5, while intermittent kisspeptin-10 infusions over 12 days sustained gonadotropin rises 3.
- FDA compounding standing: kisspeptin-10 appears in 503A category 2, while kisspeptin-54 is not named in those categories 11.
Where the evidence is limited
No study has compared kisspeptin-10 and kisspeptin-54 head to head in people. Their human studies differ in population, route and purpose: healthy men and hormone physiology for kisspeptin-10 1,3, and women undergoing IVF or with hypothalamic amenorrhoea for kisspeptin-54 5,6. Both literatures come largely from a small number of UK academic groups, and the IVF trials were open-label without an active comparator 6. Differences in dosing schedule, rather than the molecules themselves, may explain the different responses to repeated administration 3,5.
