Cagrilintide + Retatrutide
Also called Cagri + Reta, Cagrilintide/Retatrutide stack
This pairing combines two investigational agents: cagrilintide (an amylin analogue) and retatrutide (a GIP, GLP-1 and glucagon receptor agonist). Neither is FDA-approved, and no human or animal study of the pair was found. Any rationale is theoretical.
Components
What has been studied
- No human study of cagrilintide with retatrutide was found. Searched: PubMed (cagrilintide AND retatrutide; retatrutide AND amylin) and ClinicalTrials.gov (cagrilintide AND retatrutide: 0 records), October 2026. PubMed hits were reviews.
Why the pairing is discussed
Mechanistic reasoning drawn from studies of each component. Theoretical unless a study above tested the pair.
- Theoretical: cagrilintide is a long-acting amylin analogue that reduced body weight as monotherapy in a phase 2 trial 1.
- Theoretical: retatrutide is a single peptide with GIP, GLP-1 and glucagon receptor agonism; in obese mice, weight loss was attributed to GCGR-mediated energy expenditure added to GIPR- and GLP-1R-driven intake reduction 2. The pairing extrapolates the amylin-plus-incretin approach tested with semaglutide 3.
Concerns and unknowns
- Both agents cause dose-related gastrointestinal events on their own: cagrilintide in its phase 2 trial 1, and retatrutide in its phase 2 obesity trial, where gastrointestinal events were dose-related and dose-dependent heart-rate increases peaked at 24 weeks 4. Combined effects are unmeasured.
- No interaction, pharmacokinetic or safety data exist for the pair, and research-market products are not the trial materials.
- Cagrilintide has no Drugs@FDA application record 5; retatrutide is investigational and not FDA-approved.
- Neither is named on WADA's 2026 list; section S0 prohibits at all times any substance with no current governmental approval, such as drugs under clinical development 6.
