What Adipotide is
Adipotide is a synthetic peptidomimetic designed to deliver a pro-apoptotic sequence to blood vessels of white fat; the primate study that defined it reported reversible renal proximal tubule changes at the doses judged optimal, and its single registered human trial was terminated after 4 participants with no results posted.
Adipotide is a cyclic CKGGRAKDC homing motif linked by Gly-Gly to D(KLAKLAK)2 2. The homing motif was identified by in vivo phage display in mice and reported to associate with prohibitin, which the authors established as a marker of white adipose tissue vasculature and described as also expressed in blood vessels of human white fat 1. Key evidence
- Barnhart et al., Sci Transl Med · 2011 · Animals (obese rhesus macaques, baboons, cynomolgus macaques)Reduced body weight, total body fat and insulinogenic index in obese rhesus monkeys; the primary side effect was renal proximal tubule injury with glucosuria, proteinuria and creatinine rises, described as predictable and reversible at optimal amounts. 2
- Kolonin et al., Nat Med · 2004 · Animals (obese mice); human tissue for prohibitin expressionIdentified the CKGGRAKDC motif binding prohibitin on white fat vasculature; the targeted pro-apoptotic peptide ablated white fat and reversed obesity in mice, with the authors reporting no detectable adverse effects. 1
- Kim et al., Diabetes · 2012 · Animals (obese high-fat-diet mice)Glucose tolerance improved within days, independently of food intake and body-weight change, with lower serum insulin and triglycerides. 3
What the human evidence shows
Human studies of Adipotide are listed below by design, population and duration. None was designed or powered to establish an amount, and Amiravo does not reproduce their amounts for a compound approved nowhere.
- Single-arm, open-label, first-in-human phase 1 dose-escalation trial (NCT01262664), terminated per the principal investigator's request with no posted results; Men with metastatic castrate-resistant prostate cancer and obesity, no standard therapy options remaining; 4 participants; duration Study ran from May 2012 to January 2019; a single treatment cycle was planned per participant. 5
How long did human studies run?
No human study durations are recorded for Adipotide.
Cycle-length evidence for AdipotideSupplies and handling
- Lyophilized vialSealed vial with the freeze-dried cake. Check the label, lot and the certificate of analysis before opening.
- Sterile diluentThe diluent named on the product documentation. Bacteriostatic water contains benzyl alcohol; sterile water does not.
- Graduated syringesSized to the volume being measured. U-100 insulin syringes are graduated in units where 100 units = 1 mL.
- Alcohol prep padsFor wiping vial stoppers before each puncture.
- Nitrile glovesClean gloves for aseptic handling.
- Sharps containerPuncture-resistant container for used needles.
Storage
General research-context handling only: cysteine-containing peptides are sensitive to oxidation, and lyophilized material is generally kept dry and protected from light and moisture. Defer to supplier documentation for the specific lot. No temperatures, concentrations or preparation steps are given here.
- Lyophilized (unmixed) vials: typically stored cold and dry, away from light, as the documentation directs. Many are stable longer frozen.
- Reconstituted vials: typically refrigerated (about 2–8 °C), protected from light and not frozen.
- Bacteriostatic water contains benzyl alcohol, which inhibits bacterial growth in multi-use vials; sterile water has no preservative.
Handling mistakes and troubleshooting
- Added the wrong volume of diluent
- The concentration changes, not the amount of compound in the vial. Recalculate with the volume actually added and relabel the vial.
- Shook the vial and it foamed
- Let it settle in the refrigerator. Foam is a sign of agitation; some peptides aggregate when shaken. Note it on the label.
- The solution is cloudy or has particles
- Do not use it. Cloudiness can indicate incomplete dissolution, contamination or degradation. Record the lot and contact the supplier with the certificate of analysis.
- The powder cake looks melted or shrunken before reconstitution
- This can indicate the vial was exposed to heat or moisture. Record the lot and condition on arrival.
- Liquid sprayed back when the needle was removed
- Pressure built up in the vial. Withdraw a small volume of air while adding diluent to keep pressure neutral.
Regulatory status
- Status
- Not approved anywhere in the sources reviewed; no Drugs@FDA application record was returned for adipotide 7. The defining primate study reported that monkeys at the doses the authors judged optimal showed predictable and reversible changes in renal proximal tubule function 2. The only registered human study, phase 1 trial NCT01262664 at MD Anderson in men with metastatic castrate-resistant prostate cancer and obesity, began in 2012, enrolled 4 participants and was terminated per the principal investigator's request, with no results posted 5.
- United States (FDA)
- Not approved. An openFDA Drugs@FDA query for adipotide returned no application record on 24 October 2026 7. The only registered human study, phase 1 trial NCT01262664, was terminated in 2019 with no results posted 5.
- Sport (WADA)
- Not named individually in the sources reviewed; S0 covers substances without governmental approval for human therapeutic use, prohibited at all times 8.
