What Argireline is
Argireline is the trade name of acetyl hexapeptide-8, a cosmetic ingredient modelled on part of the SNAP-25 protein and marketed as a topical alternative to botulinum toxin; the small human studies located are mixed, including a double-blind split-face study that found no significant difference from the vehicle, and the proposed neuromuscular mechanism has not been demonstrated for it.
Acetyl hexapeptide-8 is a six-residue peptide, Ac-Glu-Glu-Met-Gln-Arg-Arg-NH2, whose sequence corresponds to part of the N-terminal region of SNAP-25 8. The commercial rationale is that it competes within the SNARE complex that mediates vesicular acetylcholine release at the neuromuscular junction, so that topical application reduces contraction of the mimetic muscles producing expression lines 2,6. Key evidence
- Henseler, GMS Interdiscip Plast Reconstr Surg DGPW · 2023 · Humans (19 women)Wrinkle scores fell slightly on both sides with no statistically significant difference between the side receiving the peptide and the side receiving the serum alone; the author concluded the peptide's effect was not demonstrated. 3
- Aruan et al., J Clin Aesthet Dermatol · 2023 · Humans (21 Indonesian women aged 26 to 55 with crow's feet)Improvement was reported in several subjects in both peptide arms; the authors judged the palmitoyl pentapeptide-4 arm to show more change than acetyl hexapeptide-3 and placebo, and described the study as preliminary. 4
- Acuner et al., Aesthetic Plast Surg · 2026 · Animals (40 female Wistar rats, McFarlane flap model)The peptide alone produced a collagen type I/III profile indistinguishable from saline control and no detectable tissue-level effect; flap survival did not differ between groups. 5
What the human evidence shows
Human studies of Argireline are listed below by design, population and duration. None was designed or powered to establish an amount, and Amiravo does not reproduce their amounts for a compound approved nowhere.
- Double-blind, split-face comparison of a hyaluronic acid serum with and without the peptide, instrument-measured wrinkle scores; Women recruited from a plastic surgery clinic; 19 participants; duration 4 weeks. 3
- Double-blind randomized three-arm trial (acetyl hexapeptide-3 cream, palmitoyl pentapeptide-4 cream, placebo) with corneometry, tewametry, cutometry, photography and a grading scale; Indonesian women aged 26 to 55 with crow's feet; 21 participants; duration 8 weeks. 4
- Ex vivo plus clinical study of a multi-ingredient serum (peptide combined with dipeptide diaminobutyroyl benzylamide diacetate and gluconolactone), so no effect is attributable to the peptide alone; Not isolated to the peptide; duration Not isolated to the peptide. 12
How long did human studies run?
No human study durations are recorded for Argireline.
Cycle-length evidence for ArgirelineSupplies and handling
- Lyophilized vialSealed vial with the freeze-dried cake. Check the label, lot and the certificate of analysis before opening.
- Sterile diluentThe diluent named on the product documentation. Bacteriostatic water contains benzyl alcohol; sterile water does not.
- Graduated syringesSized to the volume being measured. U-100 insulin syringes are graduated in units where 100 units = 1 mL.
- Alcohol prep padsFor wiping vial stoppers before each puncture.
- Nitrile glovesClean gloves for aseptic handling.
- Sharps containerPuncture-resistant container for used needles.
Storage
General research-context handling only. Lyophilised peptides of this type are generally kept dry, cool and protected from light, moisture and oxidising conditions, with methionine-containing sequences particularly oxidation-sensitive; aqueous solutions are treated as short-lived. Finished cosmetic formulations follow the manufacturer's own stability data. Defer to supplier documentation for the specific lot.
- Lyophilized (unmixed) vials: typically stored cold and dry, away from light, as the documentation directs. Many are stable longer frozen.
- Reconstituted vials: typically refrigerated (about 2–8 °C), protected from light and not frozen.
- Bacteriostatic water contains benzyl alcohol, which inhibits bacterial growth in multi-use vials; sterile water has no preservative.
Handling mistakes and troubleshooting
- Added the wrong volume of diluent
- The concentration changes, not the amount of compound in the vial. Recalculate with the volume actually added and relabel the vial.
- Shook the vial and it foamed
- Let it settle in the refrigerator. Foam is a sign of agitation; some peptides aggregate when shaken. Note it on the label.
- The solution is cloudy or has particles
- Do not use it. Cloudiness can indicate incomplete dissolution, contamination or degradation. Record the lot and contact the supplier with the certificate of analysis.
- The powder cake looks melted or shrunken before reconstitution
- This can indicate the vial was exposed to heat or moisture. Record the lot and condition on arrival.
- Liquid sprayed back when the needle was removed
- Pressure built up in the vial. Withdraw a small volume of air while adding diluent to keep pressure neutral.
Regulatory status
- Status
- A cosmetic ingredient, not an approved drug. Cosmetic ingredients in the United States are not subject to FDA premarket approval, and acetyl hexapeptide does not appear in any of the three categories of FDA's 503A bulk drug substances list as updated May 14, 2026 9. The Expert Panel for Cosmetic Ingredient Safety concluded in 2025 that acetyl hexapeptide-8 amide raised no safety concern in cosmetics at concentrations up to 0.005% as currently used, and that available data are insufficient to reach that conclusion above that concentration 1.
- United States (FDA)
- Marketed as a cosmetic ingredient rather than a drug. Cosmetic ingredients in the United States are not subject to FDA premarket approval, and acetyl hexapeptide does not appear in any of the three categories of FDA's 503A bulk drug substances list as updated May 14, 2026 9. Absence from drug databases is therefore expected and says nothing about efficacy.
- Sport (WADA)
- Not named individually in the sources reviewed; S0 covers substances without governmental approval for human therapeutic use, prohibited at all times 10. Cosmetic status does not by itself place a substance outside the List, since the List is organised by substance rather than product category; athletes should seek a ruling from their anti-doping organisation.
Status varies by country and changes over time. How we describe regulatory status.
