What DSIP is
DSIP is a naturally occurring neuropeptide first isolated from rabbit brain, investigated in sleep, stress and neuroendocrine research.
No receptor, gene or signalling pathway for DSIP has been established. The peptide was isolated on the basis of a biological activity rather than a molecular target, and a 2006 review states that the link between DSIP and sleep has never been further characterised, in part because the DSIP gene, protein and any related receptor were never isolated 5. Key evidence
- Schneider-Helmert, Eur Neurol · 1987 · Humans (14 middle-aged chronic insomniacs)Night sleep improved with first and repeated administration and the effect persisted into the first post-treatment placebo night; daytime alertness and performance increased. A positive result in a very small sample. 7
- Bes et al., Neuropsychobiology · 1992 · Humans (16 chronic insomniacs)Sleep efficiency and sleep latency favoured DSIP, but the authors judged the effects weak and partly due to an incidental change in the placebo group; subjective sleep quality did not change, and they concluded short-term treatment was unlikely to be of major therapeutic benefit. 8
- Kovalzon and Strekalova, J Neurochem · 2006 · Review, drawing on rabbit and rat workConcluded that the DSIP-as-sleep-factor hypothesis is extremely poorly documented and still weak, noting that no DSIP gene, protein or receptor has been isolated and that artificial analogues rather than DSIP itself promoted slow-wave sleep in their earlier animal studies. 5
What the human evidence shows
We searched PubMed and ClinicalTrials.gov and found no human studies that administered DSIP. The research described on its page is from cells or animals, and animal amounts do not translate to people.
How long did human studies run?
No human study durations are recorded for DSIP.

Supplies and handling
- Lyophilized vialSealed vial with the freeze-dried cake. Check the label, lot and the certificate of analysis before opening.
- Sterile diluentThe diluent named on the product documentation. Bacteriostatic water contains benzyl alcohol; sterile water does not.
- Graduated syringesSized to the volume being measured. U-100 insulin syringes are graduated in units where 100 units = 1 mL.
- Alcohol prep padsFor wiping vial stoppers before each puncture.
- Nitrile glovesClean gloves for aseptic handling.
- Sharps containerPuncture-resistant container for used needles.
Storage
General research-context handling only. Lyophilised peptide is normally kept cold, dry and shielded from light, and tryptophan-containing sequences are particularly light- and oxidation-sensitive once dissolved. Defer to the storage temperature, retest date and solvent guidance in the supplier's documentation for the specific lot. No preparation or handling instructions are given here.
- Lyophilized (unmixed) vials: typically stored cold and dry, away from light, as the documentation directs. Many are stable longer frozen.
- Reconstituted vials: typically refrigerated (about 2–8 °C), protected from light and not frozen.
- Bacteriostatic water contains benzyl alcohol, which inhibits bacterial growth in multi-use vials; sterile water has no preservative.
Handling mistakes and troubleshooting
- Added the wrong volume of diluent
- The concentration changes, not the amount of compound in the vial. Recalculate with the volume actually added and relabel the vial.
- Shook the vial and it foamed
- Let it settle in the refrigerator. Foam is a sign of agitation; some peptides aggregate when shaken. Note it on the label.
- The solution is cloudy or has particles
- Do not use it. Cloudiness can indicate incomplete dissolution, contamination or degradation. Record the lot and contact the supplier with the certificate of analysis.
- The powder cake looks melted or shrunken before reconstitution
- This can indicate the vial was exposed to heat or moisture. Record the lot and condition on arrival.
- Liquid sprayed back when the needle was removed
- Pressure built up in the vial. Withdraw a small volume of air while adding diluent to keep pressure neutral.
Regulatory status
- Status
- DSIP (delta sleep-inducing peptide), referred to by FDA as emideltide, has no approved drug application in the United States; Drugs@FDA / openFDA searches return no records 1. Emideltide-related bulk drug substances were on the agenda of FDA's Pharmacy Compounding Advisory Committee meeting of July 23-24, 2026, where FDA proposed that emideltide (free base) and emideltide acetate NOT be included on the 503A bulk drug substances list 23. DSIP is not named in the WADA 2026 Prohibited List 4.
- United States (FDA)
- Not approved. Drugs@FDA / openFDA returns no application for emideltide or for delta sleep-inducing peptide 1. The substance was taken to the Pharmacy Compounding Advisory Committee on July 24, 2026 (morning session) for a vote on inclusion on the 503A bulks list 2, and FDA's briefing document states "FDA is proposing that Emideltide (free base) NOT be included on the 503A Bulks List" and the same for emideltide acetate; footnotes record that both nominations (LDT Health Solutions, Inc. on behalf of International Peptide Society, and Wells Pharmacy Network) were "withdrawn by the nominator" and that FDA elected to proceed with the presentation anyway 3.
- Sport (WADA)
- Not named. Full-document word searches of the WADA 2026 Prohibited List for "delta sleep", "DSIP" and "emideltide" return nothing, so DSIP is not a named prohibited substance for 2026 4. The list's class definitions are open-ended, so athletes should consult their anti-doping organisation rather than treat this as clearance.
Status varies by country and changes over time. How we describe regulatory status.
