VK2735

VK2735 is an investigational peptide that activates both the GLP-1 and GIP receptors and is being studied as a once-weekly injection and as an oral formulation. One 13-week phase 2 randomized trial has been published, and phase 3 trials are under way. It is not an approved medicine, and this entry is for research information only.

Key takeaways

  • In the 13-week VENTURE phase 2 trial, mean body-weight change ranged from -9.1% (2.5 mg) to -14.7% (15 mg) with weekly VK2735 versus -1.7% with placebo 1.
  • At least 5% body-weight reduction was reported in 93% of VK2735 recipients versus 12% with placebo 1.
  • Gastrointestinal adverse events affected 51-83% of VK2735 groups versus 37% with placebo, and the authors reported that they became less frequent after titration 1.
  • VENTURE was short, sponsor-funded and not powered for most between-group comparisons, and the receptor-binding data it cites are unpublished company data 1.
  • VK2735 is not FDA-approved; 78-week phase 3 trials in people with and without type 2 diabetes are registered as active 5,6,9.

Development stage

Where VK2735 sits between laboratory work and regulatory approval.

  1. Preclinical
  2. Phase 1
  3. Phase 2
  4. Phase 3 (current)
  5. Approved

Stage comes from the regulatory record and ClinicalTrials.gov studies naming the compound (for compounds approved nowhere, only completed trials or trials with posted results count) (checked 2026-10-24). A registered trial is not a result.

Mechanism of action

VK2735 is described as a GLP-1 receptor and GIP receptor dual agonist that incorporates the activities of both incretin hormones 1.

Receptor binding

The VENTURE report states that VK2735 binds with high affinity to the GLP-1 receptor (IC50 188 nM) and the GIP receptor (IC50 325 nM), citing unpublished data from the sponsor 1. No independent peer-reviewed pharmacology paper was located for this entry 1.

Half-life extension and oral development

The peptide contains a side chain that imparts an extended half-life when given by subcutaneous injection 1. The developer has also registered a 13-week oral dose-finding trial in which VK2735 is described as a peptide GLP-1 and GIP dual agonist given daily 4.

Research overview

Human research

The only peer-reviewed human efficacy report located is the 13-week phase 2 VENTURE trial of weekly subcutaneous VK2735 in 176 adults with obesity or overweight 1. A phase 1 study 3, an oral phase 2 study 4 and four phase 3 trials 5,6,7,8 are registered, without posted results for those reviewed.

Animal research

The VENTURE report describes preclinical findings only as unpublished sponsor data, and no peer-reviewed animal study was located for this entry 1.

In-vitro research

Binding IC50 values of 188 nM (GLP-1 receptor) and 325 nM (GIP receptor) are cited in the VENTURE report as unpublished sponsor data 1.

Emerging safety signals

  • One participant had atrial fibrillation (judged possibly related) and dehydration (judged probably related), which led to study discontinuation; the results text places this participant in the 5.0 mg group, while the corresponding adverse-event table footnote sits in the 15 mg column, so the dose group is uncertain 1. 1 (2026)
  • Mean heart rate rose from baseline by 3.3 to 9.3 beats per minute across VK2735 groups versus 1.4 with placebo in the trial's cardiometabolic table; the trial was 13 weeks long with about 35 participants per group 1. 1 (2026)
  • One death (head trauma from a fall) occurred in the 15 mg group and was assessed as not related to the study drug 1. 1 (2026)

What is not known: No regulatory approval exists, so there is no label. The elimination half-life and peer-reviewed preclinical pharmacology were not located. The only published efficacy trial lasted 13 weeks and was not powered for most comparisons between groups. Results of the oral phase 2 trial were not posted on the registry record or published in the sources reviewed. Long-term efficacy, safety and outcome data await the 78-week phase 3 trials. The published trial was funded by the developer, and several authors are company employees.

Quality and testing notes

No PubChem compound record for VK2735 was returned on 22 October 2026, and its structure has not been published in the sources reviewed, so a reference molecular weight could not be verified [11]. A certificate of analysis for a VK2735 research material would be expected to show HPLC purity, mass-spectrometry identity, endotoxin and net peptide content; without a public structure, identity claims cannot be checked against an independent reference [1, 11].

Storage and handling

Storage conditions for research material are set by the supplier's documentation; lyophilized peptides are generally sensitive to heat, light and moisture.
